Cancer drug reverses Alzheimer's disease in mice: Hope for humans?
A new study of a promising Alzheimer's treatment has doctors buzzing that the drug may reverse the deadly neurodegenerative disease. But the new treatment isn't a new drug at all, rather a skin cancer pill that's been FDA-approved for more than a decade.
The drug, bexarotene, reversed signs of Alzheimer's in mice brains and also improved their memory in as little as 72 hours, according to the study.
"This is an unprecedented finding," study author Paige Cramer, a PhD candidate at Case Western Reserve School of Medicine, said in a university written statement. "Previously, the best existing treatment for Alzheimer's disease in mice required several months to reduce plaque in the brain."
Bexarotene - sold under the brand name Targretin - was approved by the FDA in 1999 for treating cutaneous T-cell lymphoma, a cancer of the white blood cells that primarily affects the skin. But the drug also happens to switch on a gene that makes a naturally occurring protein in the brain called Apolipoprotein E, or ApoE.
Previous research by study co-author Dr. Gary Landreth, a professor of neurosciences at Case Western Reserve, showed that ApoE could help facilitate the removal of amyloid beta proteins, which are a toxic substance in the brain that causes plaque buildup that's a marker for Alzheimer's.
Dr. Landreth and his grad student, Cramer, thought if they could use this drug to increase ApoE, then maybe they could reduce some of the plaques in mice that were genetically bred to have Alzheimer's. The researchers found that the drug caused a 50 percent reduction in the amount of amyloid plaques within only three days, and after 14 days, they saw a 75 percent reduction. The drug essentially "reprogrammed" the brain's immune cells to "eat" the amyloid plaques.
The brain plaque reduction was also associated with improvements in the rat's behavior. Mice instinctively make nests out of materials around them, but mice with Alzheimer's typically fail this task. But after 72 hours of bexarotene treatment, the Alzheimer's mice could build nests from tissue paper, and also performed better on other tests, which suggest a "clearer thought process," Cramer said in a university video.
The study is published in the Feb. 9 issue of the journal, Science.
One caveat to the study - mice aren't people. Despite how promising the findings may seem, the researchers tempered expectations.
"I want to say as loudly and clearly as possible that this was a study in mice, not in humans," Landreth told CNN. "We've fixed Alzheimer's in mice lots of times, so we need to move forward expeditiously but cautiously."
Dr. Maria C. Carrillo, director of scientific relations at the Alzheimer's Association agreed and told the Washington Post, "We need to be cautiously optimistic and pursue this lead as we would any other." But she added, "This an exciting study is that it involves a repurposed drug."
The researchers hope to get approval to study bexarotene in humans as soon as possible, and Landreth told the Post there's an urgency since he's heard from other doctors that patients are already asking about bexarotene.
"We've got to work fast, and we have got to be right," Landreth said. "We can't screw this up."
Alzheimer's is the sixth leading cause of death in the U.S., according to the CDC, taking more than 83,000 lives this past year. “Whether it’s an obsession or an addiction, I don’t know. It’s certainly different from a few years ago, partly because the technology wasn’t there,” he said.
Robert Hart Fletcher, who runs Kids and Media, a charity giving information about children’s use of digital media, stated “Gaming is a phenomenon that’s been around quite a while.Now we are starting to see the effects in behaviour of young people.”

"Simple-to-prepare, healthy breakfasts for children can include fresh fruit and high-fiber, lower-sugar cereals. Better yet, pair that fruit with homemade oatmeal."
Earlier this year, the federal panel proposed voluntary guidelines for foods nutritious enough to be marketed to children, including limits on sugar (no more than 26% by weight), sodium (no more than 210 mg by 2016 and 140 mg by 2021), and fats (no more than 1 gram of saturated fat per 50 to 55 grams of cereal), and a target for more than 50% whole-grain content.
Overall, 67% have too much sugar, 12% have too much sodium for the initial standard and 85% have too much for the final standard, 8% have too much saturated fat, and at least 31% do not have enough whole grains.
"The proposed federal guidelines are more restrictive than those of the food industry, but they should be strengthened and made mandatory," the advocacy group concluded.
In reference to Cap'n Crunch, which EWG had at number five on the list of cereals with the highest sugar content, the company said, "While it's not marketed as a health product, it does include essential vitamins and minerals and offers consumers a low-fat breakfast option when paired with low-fat milk and a serving of fruit."

EWG also provided a list of big-brand cereals (both children's and nonchildren's) that meet all of the federal nutrition guidelines and can be found easily, but may contain ingredients that are genetically modified or grown with pesticides:
- Kellogg's Mini-Wheats: Unfrosted Bite-Size, Frosted Big Bite, Frosted Bite-Size, Frosted Little Bite
- General Mills Cheerios Original and Kix Original (these two meet the 2016 sodium guideline but not the final guideline for 2021)
- Post Shredded Wheat (all varieties), Grape-Nuts Flakes, Bran Flakes, and Honey Bunches of Oats with Vanilla Bunches (the latter three meet the 2016 sodium guideline but not the final guideline)
- Quaker Oats Oatmeal Squares Cinnamon (this meets the 2016 sodium guideline but not the final guideline)
Cereal makers have opposed the federal recommendations, and have created their own set of standards, which were published this year by the Better Business Bureau's Children's Food and Beverage Advertising Initiative and are scheduled to be implemented in 2014.
According to these standards, cereals can contain up to 38% sugar by weight.
The EWG pointed out that 21 of the cereals included in its study would not meet the industry's proposed guideline.
"The proposed federal guidelines are more restrictive than those of the food industry, but they should be strengthened and made mandatory," the advocacy group concluded.
In an emailed statement, Quaker Oats said, "We offer consumers a wide range of wholesome breakfast options, including heart-healthy Quaker oatmeal as well as Quaker Lower Sugar Instant Oatmeal, and we adhere to marketing standards that prohibit advertising any products to children under 12 unless they meet specific nutrition criteria."
In reference to Cap'n Crunch, which EWG had at number five on the list of cereals with the highest sugar content, the company said, "While it's not marketed as a health product, it does include essential vitamins and minerals and offers consumers a low-fat breakfast option when paired with low-fat milk and a serving of fruit."

Video games 'don't make kids violent': study
While children who play violent computer games are more aggressive, there's no evidence to support claims that the games themselves cause kids' aggressive behaviour, the Swedish Media Council (Statens medierĂ¥d) has concluded.
The findings come following a review more than 100 articles about violent computer games and aggression which have been published in international scientific journals since 2000.
According to the Council's review, there is a clear, statistically significant link between violent computer games and aggressive behaviour.
However, many of the studies use different methods to measure aggression, many of which lack a clear connection to violent behaviour.
In addition, a great deal of the research exploring causal links between violent computer games and aggressive behaviour “suffer from serious methodological deficiencies” and don't provide sufficient evidence to establish a causal relationship.
The few studies that have attempted to examine other causes of aggression found that factors such as poor physical health or family problems can explain both violent behaviour and a propensity to play violent computer games.
There is also too little research into the hypothesis that violent video games may cause violent behaviour among people with certain personality traits.
As a result, the Council concludes, “there is no evidence that violent computer games cause aggressive behaviour”.
“If research can't provide any simple answers about how games make children aggressive, perhaps we adults should stop judging the games children play based on whether they are violent or not,” Media Council researcher Ulf Dalquist said in a statement.
Speaking with the Aftonbladet newspaper Dalquist said that a number of longitudinal studies included in the review showed “beyond a reasonable doubt” that family conflicts and poor health among children can make children more drawn to computer games and result in a higher level of aggression.
And while the games themselves may not cause aggressive behaviour among children, parents should nevertheless be aware of the games their children play.
“As parents, we can stand and freeze on the sidelines of a football pitch or listen to out of tune recorder concerts – but we can't engage in their computer gaming. It may not be about how much violence a game contains, but rather what sort of values the game expresses,” he told the newspaper.more information...

Local Internet Marketing Company Ajax Union Uses Facebook to Join Fight Against Breast Cancer
In affiliation with Google and the Susan G. Komen for the Cure’s Pink Pin Initiative, search engine agency Ajax Union has developed a Facebook campaign to raise funds for and awareness about the growing presence of breast cancer in the United States.
In an effort to promote awareness about the rise in cases of breast cancer in the United States, Ajax Union - a business marketing company online at http://www.AjaxUnion.com - has taken part in the Pink Pin Initiative, a partnership between Google and breast cancer awareness group Susan G. Komen for the Cure.
According to Breastcancer.org, approximately 1 in 8 U.S. women will develop breast cancer in the course of their lifetimes. To raise awareness of this staggering statistic, Ajax Union created "1 in 8," a Facebook campaign in which the internet marketing company will donate $1 for each new fan its Facebook page receives before the end of November, as well as an extra $1 for every eighth fan, up to $100.
“People don’t always realize just how widely the effects of breast cancer can reach. Anyone could develop it - a relative, a friend, a neighbor. It doesn’t discriminate. We heard about the Pink Pink Initiative and thought that would be the perfect way to do our part to help,” said Sarah Mogin, head of social online marketing and PR at Ajax Union.
Breastcancer.org also reports that over the course of 2011, there have been over 2.6 million breast cancer survivors in America. Ajax Union is hoping to do its part to raise that number.
"Being one of the first 1,000 New York businesses to register for the Google Pink Pin cause, we really wanted to inspire and rally everyone in our network to join the fight against breast cancer," said Elizabeth Apfelbaum, a Vice President at the internet marketing company.
As seen on the Pink Pin map at http://www.PinkPin.com, the majority of participating businesses are located in New York City, although companies throughout the country have also stepped up to participate. A handful of businesses are participating in other countries as well, including Mexico, Chile, and Saudi Arabia. In the first 24 hours alone 300 businesses signed up to take part in the Pink Pin Initiative.
Registered companies have an extra incentive to raise funds - Google has promised to match up to $50,000 of all donations made by Ajax Union SEO NY and the other participating business, bringing the projected grand total to $100,000.
